Matheson was born in Glendale, a suburb of Los Angeles. At the age of 13, Matheson appeared as Roddy Miller in the comic series Ventana on Main Street (CBS) with actor Robert Young during the 1961-1962 television season. In 1964 he provided the voice of the main character of the cartoon show Jonny Quest. It was also the voice of Jason in the original animated series Space Ghost. He also played the role of the eldest son, Mike Beardsley, in the film Mine, yours and ours, starring also Lucille Ball and Henry Fonda. During the filming of this film he met the Canadian actress Jennifer Leak (1950-), with whom he married.
In 1969 he joined the NBC cast The Virginian (Western series) in the eighth season, as Jim Furnaces. He had a guest role in episode 14 of the second season of Night Gallery, in the chapter «Cabezas de Logoda». In the last season of the western Bonanza (in 1972-1973), he played Griff King, a prisoner on parole who tries to reform his life as a worker at Rancho Ponderosa under the watchful eye of Ben Cartwright. He played a young motorcyclist cop, Phil Sweet, in the 1973 movie Magnum Force. Matheson also appeared early in the comedy TV series My Three Sons and Leave It to Beaver (on ABC).
In the fall of 1976, Matheson appeared with Kurt Russell in the 15-episode NBC episode The Quest, the story of two young men in the American West searching for the whereabouts of their sister, a captive of the Cheyenne. In 1978 he was co-star in Animal House with John Belushi. The following year he appeared again with Belushi in 1941 (by Steven Spielberg). Matheson starred in the comedy film Hasta el arroyo (from 1984) and the comedy Fletch.
Matheson also appeared in Soy o no soy (1983), starring Mel Brooks and Anne Bancroft.
Matheson and Catherine Hicks represented Rick and Amanda Tucker, who operate a detective agency in Laurel Canyon, (Los Angeles) on Tucker’s Witch (CBS), which aired during the 1982-1983 season.
Matheson, along with his business partner Grodnik Dan, bought National Lampoon’s Van Wilder magazine in 1989, when the magazine was in financial decline. They were unable to reverse the fate of the magazine, however, and sold it in 1991. Matheson went on to act in more than 100 films and television projects. He had a recurring role as Vice President Juan Hoynes in The West Wing. That work earned him two Emmy nominations as “best guest actor in dramatic series.” In addition to playing Sheriff Matthew Donner on Wolf Lake, he has directed episodes of Third Watch, Ed, The Twilight Zone, Cold Case, Without a Trace, The West Wing, Burn Notice, Psych, Shark, White Collar and Criminal Minds.
In 1996, Matheson assumed the role of a swindler who claims to be the missing husband of Carol Brady in A Very Brady Sequel. Matheson appeared in the movie Van Wilder in 2002, playing the father of the protagonist, inspired by his own character in Animal House; even the character of Matheson makes a veiled reference to the old character. He appeared in the Redline racing movie. He also appeared in a Volkswagen commercial in 2008.
In 2009, Matheson directed the pilot episode of Covert Affairs, which premiered on the USA Network in 2010.
He later directed the Fox television series The Good Guys, in Dallas, Texas.
After separating from his first wife, he is married to Megan Murphy Matheson, with whom he has three children.
